For the sake of discussion, Bob is completely innocent, and unaware of anything related to the following conversation. It involves an accusation by Bill against Bob.
Bill: Bob killed the accountant and stole a million dollars from the company!
Bob: I did not! Are you crazy?
This is the wrong way to respond. It dignifies the blatant lie and also opens the door for Bill to talk some more. This in not the correct approach.
The correct way to respond might be
Bill: Bob killed the accountant and stole a million dollars from the company!
Bob: You are a goddamn liar, and if you try that shit on me I will fucking bury you. Also I will kill your entire family, including your fucking dog. Don't say a fucking word because I know you heard me asshole.
This approach puts Bill on notice that his tomfoolery will not be tolerated, and also that he will die if he persists.
The correct way to respond

Yes, but one learns the hard way not to announce such things publicly

Re: The correct way to respond
There is something to what you said here. The way you handle an accusation controls how your opponent is able to proceed; but rambling about murdering peoples family's and such is probably not a good way of going about it imo..

Re: The correct way to respond
After reflecting on it all, I now think Bill was the one who killed the accountant and stole all the money
